A TIRF(Total Internal Reflection Fluorescence) microscopy system utilizes evanescent wave illumination. This illumination causes an extremely thin section of fluorophor excitation (typically less than 100nm) resuting in high-contrast real time imaging of labeled molecules at the cell membrane or basal plane of the specimen.
The unique extendible configuration of the Nikon TE2000 microscope enables simultaneous mounting of both TIRF and epi-fluorescence illumination systems, which allows easy switching between the two, or image overlay.
The dedicated TIRF objective(60X 1.45N.AA)has a correction collar for cover-glass thickness calibrated at 23℃(room temperature) or 37℃ (physiological temperature). This feature allows for correction in temperature-induced changes in refractive index of immersion oil that could induce spherical aberration. |
